Inhibition of urease by copper salts in the presence of ascorbic acid and related substances.

چکیده
Elson (1943) found that urease is inhibited by small amounts of ascorbic acid and that this inhibition is eliminated in the presence of cysteine. Quastel (1943) suggested that the inhibition was due to a reaction between dehydroascorbic acid and an -SH group of the enzyme, and that the protective effect of thiol compoundcls was due to the fact that they reduced dehydroascorbic acid. Giri & Seshagiri Rao (1944), however, did not find that dehydroascorbic acid inhibited urease, and since they observed that there was no inhibition of the enzyme by ascorbic acid in the presence of substances which form complexes with copper (8-hydroxyquinoline and diethyl-dithio-carbamate), they suggested that the inhibition might be due to the formation of intermediary substances like Cu2O, produced during the oxidation of the vitamin. Our experiments show that neither ascorbic, nor its oxidation products, dehydroascorbic acid or 2:3-diketo-gulonic acid, themselves inhibit urease. Ascorbic acid and 2:3-diketo-gulonic acid, however (but not dehydroascorbic acid), act as inhibitors in the presence of Cu++. Cu+ inhibits urease to a much greater extent than does Cu++, and the increased inhibition observed when ascorbic acid is added to a solution of the enzyme containing Cu++ is due, it is therefore concluded, to the reduction of Cu++ to Cu+.

Determination of L- Ascorbic Acid in Plasma by Voltammetric Method
Voltammetric techniques have been considered as important methods among the analytical techniques used for the identification and determination of trace concentrations of many biological molecules such as L-ascorbic acid (AA). L-ascorbic acid is an electro-active molecule, though it is difficult to determine its value directly with a majority of electrodes made of carbon and transition metals, ...

متن کاملAlleviation of Adverse Effects of Copper on Allium cepa L. by Exogenous Ascorbic Acid Application
Abstract In this study ameliorative effects of ascorbic acid (0.5 mM) on onion (Allium cepa L. cv. Red Azarshahr) under copper stress (Cu: 6.5 μM) were studied. Fresh weights of leaves and roots were reduced in seven week old plants under excess copper condition. Cu stress also reduced membrane stability index in leaves. There was a considerable increase in H2O2 content and lipid peroxidation i...
متن کاملAscorbic acid augments colony spreading by reducing biofilm formation of methicillin resistant Staphylococcus aureus
Objective(s):Staphylococcus aureus is a Gram-positive pathogen, well known for its resistance andversatile lifestyle. Under unfavourable conditions, it adapts biofilm mode of growth. For staphylococcal biofilm formation, production of extracellular polymeric substances (EPS) is a pre-requisite, which is regulated by ica operon-encoded enzymes. This study was designed to know the impact of ascor...
متن کاملThe Effect of Polyvinylpyrrolidone on the Formation of Copper Nanoplates in Wet-Chemical Reduction Method
In this work, we report synthesis and characterization of copper nanoparticles in polymer matrix by wet-chemical reduction method using ascorbic acid as reducing agent, copper (II) sulfate as metal precursor and polyvinylpyrrolidone k-30 (PVP K-30) as surfactant agent. The reaction was carried out in a high-speed stirring mixture at room temperature under nitrogen atmosphere. Characterization o...
